Kelun-Biotech Advances First Dual-Payload ADC into Clinical Trials
Sichuan Kelun-Biotech has secured approval from China’s Center for Drug Evaluation to begin human testing of SKB565, the firm’s first antibody-drug conjugate to utilize a dual-payload mechanism. The drug, designed to treat advanced solid tumors, targets cancer cells while simultaneously triggering an immune response within the tumor microenvironment.

Developed on the company’s proprietary OptiDC™ platform, SKB565 moves beyond traditional single-payload therapies by delivering both toxins and immunomodulators directly to diseased tissue. This combination approach aims to overcome common drug resistance patterns by attacking tumors through two complementary pathways. Preclinical data indicated the molecule possesses robust antitumor activity, clearing the path for the current regulatory milestone.
CEO Dr. Michael Ge described the candidate as a cornerstone of the firm’s “ADC + IO” strategy, which seeks to integrate synergistic functions into a single molecule. The company is currently managing over 30 innovative drug projects, with a heavy emphasis on solid tumors and metabolic diseases.
