Enliven Therapeutics Secures FDA Alignment for ELVN-001 Phase 3 Trial

Enliven Therapeutics has cleared a critical regulatory hurdle for its lead chronic myeloid leukemia candidate, ELVN-001, securing FDA alignment on Phase 3 trial design. The Burlingame-based firm, bolstered by an $895 million cash reserve, is now preparing to launch its ENABLE-2 study in the second half of 2026.

The company’s clinical outlook follows positive Phase 1 data presented at the European Hematology Association Congress. In the 80 mg once-daily cohort, ELVN-001 achieved a 61% overall major molecular response, with efficacy metrics proving particularly robust among patients with limited prior exposure to tyrosine kinase inhibitors. The FDA has granted the drug Fast Track Designation, further accelerating its path toward potential commercialization.

CEO Rick Fair noted that the recent clinical milestones and the strengthened balance sheet position the company to advance its precision medicine pipeline rapidly. While research and development expenses rose to $29 million for the second quarter of 2026, the company’s current liquidity is projected to support operations through 2030. Final design components for the ENABLE-2 trial are expected to be solidified following an anticipated End-of-Phase 2 meeting later this year.
