Phanes Therapeutics Begins Colorectal Cancer Dosing for Spevatamig
A patient with colorectal cancer has received the first dose of spevatamig, marking a new milestone in the TWINPEAK clinical trial. This study evaluates the efficacy of the bispecific antibody, designed to target both claudin 18.2 and CD47, in patients suffering from advanced gastrointestinal malignancies.

Spevatamig represents a novel class of immuno-oncology agents known as innate immunity enhancers. By targeting claudin 18.2 and CD47, the drug aims to address high unmet medical needs across various solid tumors. Its potential is currently under scrutiny in the multi-center Phase I/II TWINPEAK study, which monitors safety, tolerability, and pharmacokinetics in cases of gastric, pancreatic, and biliary tract adenocarcinomas alongside colorectal cancer.
The development program has already garnered significant regulatory attention, securing FDA orphan drug and Fast Track designations for pancreatic and biliary tract cancers. Furthermore, Phanes Therapeutics is testing the antibody’s versatility through a clinical collaboration with Merck, exploring its effectiveness when paired with pembrolizumab. Beyond the U.S.-based TWINPEAK trial, the company is actively recruiting for additional clinical phases in China to broaden the scope of its gastrointestinal cancer research.
